My only words of advice are to ensure you get a few views on the work before posting and creating buzz. Depending on what kind of humour you're targeting. Is it a cute-funny video? is it one where someone hurts his/herself? Try to target the largest audience possible when choosing your footage.
Also, like Mike said, the editing of the video could be crucial to it's success. By keeping it short and sweet you may find a better success rate.. Also sometimes repetition, slow motion, and adding music (if it doesn't have any yet) helps
